If you are in the lab exam and the task states "ensure all ports connecting to routers transition to forwarding as quickly as possible" - I check each switchport and ensure portfast is used with access, and portfast trunk is used with trunks (router on a stick). There would be no reason that I can think of to do both in the lab exam. Just do the appropriate one for the configuration.
