It does not require a full slot, but depending on your tunnel bandwidth requirement it may require dedicating a port to the tunnel functionality.

You can configured a 1Gbps tunnel interface on a 40 x 1GE DPC and still use all 40 1GE ports. Enabling a 10Gbps tunnel interface on a 4 x 10GE DPC means that one of the 10GE interfaces can not be used, but the other three 10GE interfaces are still usable.
