I don't really see what the problem is:

\relative c'{
c-\tweak #'font-size #-3 _\flageolet
c-\tweak #'font-size #-3 ^\flageolet
}

 \tweak only accepts `-' due to its definition.

Is there a definition of \tweak given somewhere in some .scm file?

It's defined in music-functions-init.ly (as a Scheme function). The
exact cause of the rejection of `^' and `_' is buried deeply in
lily/parser.yy -- I haven't understood it :-)
