It can be drive/file permission where the SP IIS files are located. Also, are you using custom host header and point it to 127.0.0.1? If you are then make sure you create a new DWORD value called "DisableLoopbackCheck" and set it to 1 in the following registry key HKEY_LOCAL_MACHINE\SYSTEM\CurrentControlSet\Control\Lsa.
Also check DB user/role security. Ensure that the app pool user account has access to the content DB.
