[Histonet] Unregistered HT
Hi all, I'm wondering if anyone can share a job description for an unregistered HT position. I also need to know if unregistered HT's are allowed to cut frozen sections. I know there is a limit to what they can do, especially where IHC and special staining are concerned, but I really don't know about frozens, since it's a lot like regular microtomy, except for the H&E staining of the slide. I also understand that facilities might have different descriptions, but what I'm really looking for is what CAP allows them to do. [Histonet] Losing tissue on IHC slides
Hi all, I'm looking for some troubleshooting help. In the last couple of months we've been having an increasing trend in losing tissue from our IHC slides. We use a Ventana Benchmark XT. PM is done on it quarterly. The problem seems to be with the patient tissue, not the control tissue. We use Fisherbrand Superfrost Plus slides, picking up the control by dipping the top end of the slide to avoid "double dipping". We use tap water in the waterbaths which I know could cause an issue, but that's the way it's always been done here, so there's been no change to that part of the procedure. (We did try using DI water, but it didn't make any difference. Tissue still fell off of slides.) We do use DI water to mix the bulk reagents.The control slides may sometimes have been cut for a longer than recommended time, but that part of the procedure hasn't changed either, same process as always. If we run separate slides for control and patient tissue, we get much better results, but still have some tissue loss. I can't say it's all of our tests, but it is a steadily worsening problem. I'm going through tons of reagent, and I can't seem to nail down the problem. Ventana rep said to try DI water, don't double dip, try adhesive slides (which didn't really make any difference that we could tell). [Histonet] Radiatiation Safety
Hi all, I'm trying to complete a protocol for radioactive specimen handling. I need to know if anyone has suggestions for this. We do not use a geiger counter for specimens. The radiation safety person in nuclear med says their policy is that technicium 99 has such a short half life, and that there is no need for special handling of sentinel node specimens. I just need some input. I'm not really getting anywhere with nuclear med on any other information such as prostate seeds, the actual breast biopsy that may be sent after a sentinel node procedure, etc. Our CAP inspection looms, and I need to finalize this protocol ASAP. [Histonet] Background staining on H pylori
Hi, I'm getting a lot of background staining on my HP's. We use a Ventana BenchmarkXT with ultraView DAB. The problem seems to be escalating of late, and I'm not sure why. We use Hollande's on our GI biopsies, and run them on a shorter run. Nothing has changed in the processing of the specimens, or the IHC procedure. The antibody dispenser is a newer lot, but not brand new, as we get the larger size due to the high volume of HP's we run. Any ideas? Hi, Can anyone tell me if the control blocks we use for special stains and IHC need to have the accession number of the original case they came from on them? [Histonet] IHC new reagent lots
I'm in a new position and have to get our IHC paperwork CAP ready. I'm not sure about something. When you put a new antibody dispenser (with a different lot # than previously used) on the IHC stainer, what QC needs to be done? Can the control run with a patient test serve as your QC, or does it need to be a separate slide done before you run any patient tests with that new lot #? Also, I know that CAP is no longer requiring negative controls to be run when using a biotin free detection system. Can we just stop running the negative controls, and reference the CAP change in our protocol, or do we have to perform some sort of validation? I appreciate any help.
