[Histonet] ER/PR/Her2 control

Hello Histonet!

Looking for some advice regarding ER/PR/Her2 controls.  I'm able to buy 
commercially TMA blocks containing varying levels of positivity for ER, PR, and 
Her2.  This work nicely because you can have a negative and varying levels of 
positive control as a same-slide control.  But our docs would like to score 
Ki-67 as well.  Currently, they are just doing pos/neg for Ki67.

It will take some time to collect cases and create a similar control, so I'm 
wondering what y'all are using?  Is there anything commercially available?  Or 
is everybody just collecting cases with varying degrees of positive?

That's interesting, we've been seeing warping with the Leica coverslips.  I 
wonder if there's something in common in their manufacturing.  We've been using 
a mixture of Superfrost Plus (Fisher), Basix (Fisher), and Platinum line 
(Mercedes Medical) all with good results.  The 'Basix' seems to be a good 
all-around non-charged slide that is affordable.  Hope this helps!

[Histonet] Specimen disposal bags

Hello Histonet,

I'm looking for some recommendations on biohazard bags for specimen disposal.  
Currently using Brighton 3.0 mil 44 gallon bags and they have occasional 
problems with leakage.  Obviously the formalin is removed but there is always 
some residual formalin present.  I'm willing to pay a little more for a better 
quality bag that is more leak-resistant.  Any advice?

[Histonet] TMA slides for IHC validation

I'm using TMA slides from US Biomax to assist with IHC validation.  I'm working 
on p120 on a Benchmark Ultra and BR2410 slides from Biomax.  The cases are all 
invasive lobular carcinoma, which should all stain positively, but only about 
half are staining positive.  I sent an identical slide to our reference lab and 
the staining matches my own.

Since it's the first time I've worked with TMA slides, am I missing something 
obvious?  Are the tissue cores generally prepared in the same fashion?  Any 
reasons why I'm seeing inconsistent staining of tissue that should all be 
positive?  All regular cases pulled of invasive lobular carcinoma are staining 
appropriately, just something unusual with these TMA slides.

Any advice is appreciated!  Thanks!

Just another note:  You can order unstained tissue microarrays with the 
prerequisite number of cases, both positive and negative, and stain your 
validation all on one slide.  I've done this for years and for 3 different 
validations of entire IHC platform changes, ranging from 40 to over 100 
antibodies each time.  Saves time and money.

Hello,
We've been discussing about the quantity of slides to run as a validation for 
IHC antibodies. We are governed by CLIA, and we would like to know if there is 
a set number of slides to run for a particular antibody we would like to bring 
in-house for Validation.  I think CAP requires 20 slides..?
And so we are asking if there is  a requirement with CLIA to run a certain 
number of slides, or is it up to us (the laboratory director) to decide how 
many slides to run for Validation/Verification.

Ventana/Roche bought out MTM which had the patent, so they're the only vendor 
you can get it from.  Since you're using Bond you obviously don't want it in a 
dispenser, but you can buy their manual staining kit and just use the antibody 
from that.

[Histonet] Air filter/purifier

Hello Histonet,

Does anyone have experience with an air filter/purifier for use in the lab?  
I'm looking mainly for residual toluene fumes.  Everything is always below the 
limit but I have an employee who is unusually sensitive and I'm trying to help 
out.  I found one from Fisher for an astronomical price, and I don't know if 
there's anything that really makes it different from generic air purifiers you 
can purchase anywhere.  Does anyone use anything that they like?
