Mechanical problems, contamination and dilution of the Negative Control Serum did not appear to be factors. Biocare knows about this and is meeting with Ventana to iron out the problem. Would have been nice to know about this if it was a known issue before we bought and started using the product, but better late than never. Posting this in case anyone else on Histonet is experiencing similar staining challenges using this product. As Biocare states in the product insert sheet, when used in conjunction with non-Biocare kits, results may vary. We'll keep using it and are currently waiting for advice from Biocare. We also have Ventanas. We currently use a normal goat serum as our "non-immune" serum. We are considering changing our SOP's to use the Ventana Neg Control Rabbit and Neg Control Mouse in dispensers. Linda's suggestion of Biocare's Universal Negative Control Serum is intriguing. It would seem regressive to go back to the days of delineating mouse/rabbit antisera, using separate dispensers for each, and a universal negative seems to be the way to go, especially when using universal DAB kits. Using such a reagent would mean it could be applied to each case's negative control slide. Linda or others, would such a negative control serum protocol include your most aggressive pretreatment? My co-worker previously asked about any versions of this antibody that didn't require formic acid pretreatment. I was wondering if there is experience out there using the antibody from DBS, and what the consensus is about the stability of the antibody (from any company) after being diluted. I think that besides checking Histonet archives for past discussions on this, if you keep a file of freshly cut, freshly stained control slides that you can go back to compare day-to-day results with, you will be able to pinpoint fading stains better. You may also want to have cut control slides inventory that coincides with an estimated workload-for example, have 1 months-worth of tonsil at room-temp and 2-3 months in a frost-free freezer. If you have some at both storage conditions, then you can check the staining quality versus the control section's age as you optimize antibodies. Although this is a generalization-most of the stains we've seen affected by control age/storage conditions have been nuclear-staining antibodies. We currently cut the positive control from the block as needed. I have learned that some controls do start to deteriorate over time, tonsils for example. But, is there a list, an article, SOMETHING that can guide me so that we can cut some of these controls ahead of time? This would not only save tech time, but would further the use of our controls. In cutting the block, some of the good control is wasted and some positives are hard to find. My argument is that the purchased controls work wonderfully, even after a very long time and our improved detection kits have improved so much over the years, it makes this possible. If there is no published article out there, I implore you experts to please help us in this area. Would also like to hear from the technical experts from our suppliers too!
