[Histonet] Freezing artefact

Good Afternoon all,

I have a couple of cases where my post mortem samples have been accidentally 
frozen in Neutral Buffered Formalin. Naturally this has led to pretty striking 
artefact in the tissue. I was wondering if anyone who perhaps has more 
experience of dealing with frozen tissue has any tips on minimising this 
artefact? I still have some fixed tissue but am unable to resample from the 
unfixed tissue. Thanks in advance.

[Histonet] Attwood's tips please

Hello,
I'm trying to work up an Attwood's  method but would be keen to get a guide on 
time for the differentiation step in tartrazine solution. The method I'm using 
suggests 20 minutes at the longest with regular microscope checks. I'm 
currently at about 4 hours with only slight loss of colour in the RBC's. I know 
that the tartrazine solution will work eventually as the slides I left in over 
the weekend became fully decolourised but those time scales are unworkable.

suggested that I put up a dropbox link to all of those provided. I thought that 
this sounded like a good idea so I will do this later, if anyone would rather

[Histonet] training log

Hello all,

Does anyone have a training log or competency list that they would be willing 
to share with me for an absolute beginner to hands on histology. I have 
previously trained people with limited experience but never from scratch 
(straight from university). I am keen not to miss the basic lab skills that

[Histonet] mortuary scales advice

Hello all,

I am searching for a weighing scale that will record multiple organ weights 
during autopsy and then allow display of all data at the end of the session. 
Ideally this would be a stand-alone unit rather than computer linked. Does 
anyone have experience with this sort of equipment?

Hoping someone might be able to help with this microtome issue. I have a 
problem with the block clamp on one of my Leica RM2255's. It has recently 
loosened up and it is now possible to move the clamped cassette from 
side-to-side when the clamp is shut.

When I turn the bolt on the underside of the clamp it neither tightens up nor 
loosens off so there does not seem to be a way of taking the clamp apart any 
further.



Leica have suggested that I buy a new block clamp (£292) but it just feels like 
something that could be easily fixed, perhaps with a new spring? Has anyone had 
experience with this problem?

Just wondering if anyone knows the what the 'O' in oil red O stands for.
I suspect that it stands for nothing other than O similar to the 'G' in
Orange G. One of our pathologists is looking for a tricky stain question
to annoy trainees with.

[Histonet] obesity/cardiac study

Hello histonet this is a call for any advice.

I am a purely lab-taught histotech based in the UK and have been
studying part-time to enable suitable accreditation and for my final
year project am intending to use the large back catalogue of blocks in
the forensic lab I work in to study cardiac changes associated with
obesity (this is of special interest in the West of Scotland where I
work).

My intention is to select cases where obesity is judged (by pathologist)
to be a primary or secondary factor in cause of death and where other
factors (i.e. hypertension) were not indicated in life and compare with
an age-matched cohort to asses the correlation between obesity and
cardiac changes such as degree of interstitial fibrosis.

Initially I intend to take a single case with well developed
obesity-linked cardiac changes and do a range of stains (H&E, MSB, EVG,
PTAH, a range of trichromes, PSR) to determine which stains to use in
the whole study. I would be very grateful to hear from anyone who has
undertaken similar work with advice on range of stains/techniques or
tips on papers I should read or with any other advice. Also any advice
on immuno that I could undertake.
