I am old school and prefer them dry, lol. I agree with Thomas, that shouldn't
have that affect.

Without seeing the blocks, that sounds more like cold molds being used, more
Then, whether or not the tissues are kept in a dry, hot, well, or a wet well.

Re: [Histonet] Retic help

2023-09-18 Thread Kurth, Virginia L via Histonet
Same here, we moved to a new location and the PH of the water was too acidic,
the DI water would remove the silver and leave it yellowed. Some solutions
that I was making that usually would take hours to go into solution on the stir
plate went immediately.

Good Luck!

Hi Steven,

We ran into this issue and it was the pH of the water. We were using our
sterile line from the facility but the pH was too high. We used tap and it
worked fine. This will also effect your stock solutions if you make them
yourself so be aware of that as well.

Re: [Histonet] Sections sticking to tweezers

2023-05-08 Thread Kurth, Virginia L via Histonet
You may try to turn your water bath down a few degrees and see if this helps
and what is the melting point of the paraffin you are using? Most likely a
paraffin/heat situation.
If I run into this issue, a quick fix is I rinse my forceps into the ice
water, wipe, and the sections will not stick.

Good Luck.

Can anyone tell me why my sections are sticking to my tweezers and turning into
one long string of snot when I try to separate them? I have a good pair of
curved stainless tweezers and I am trying both the front and the back and
tapping sharply while allowing the tweezers to open at the separation point. I
can separate one or two, then it sticks and pulls up 7 or 8 sections into one
long slimy string. My water bath is set at 38.5 and I am sectioning at 4-5 m.
Even when I go to 6 it still does it. Any ideas?

Make sure they sit at room temperature longer than normal and then dry them
longer. I found that cutting them thinner (3 microns) also helps.

Yes...a big problem for me, tooif you are using HIER??
Cartilage and bone are major tissue retention problems I cut Pwax sections of
mouse long bones/knee joints for anti GFP IHC I still get a % fall off but,
using manual HIER at 90C I manage to get enough sections to get good results
However, I imagine that your autoIHC stainer prob uses 90C HIER?
I find Trajan 3 series slides more reliable than Superfrost Plus slides.
Sure, I need to dry my slides for 24 hrs in an operating fume hood ( we can't
turn ours off/down) Then I place slides in a 60C oven for 2hrs.
Then dewax etc

Be interesting to read of other suggestions

Best wishes

  3. Do you have any specimen types that are processed as whole mounts as

  5. Are you aware of how radiologic/pathologic correlation is performed on
