[Histonet] Amyloid A controls

Hello Histonetters,

I am looking for a good "Amyloid A control".  We have an anti-amyloid A 
antibody which is specific only for amyloid A.  We have tried our commercial 
congo red controls and they do not work.  We have only had a few positive 
cases.  I am willing to trade control blocks.  We have parvovirus, h,pylori,  
C4d on heart, and many other controls just ask and maybe we could make a trade.

This is a well known phenomenon of amyloid staining and it is fairly 
common. It is due to the fact that amyloid is a very variable material 
and different samples may react differently to the dyes. I suggest you 
restain using Highman's congo red method 
(http://stainsfile.info/StainsFile/stain/amyloid/congohighman.htm) is 
more reliable than Bennhold's. I always found it the more reliable. If 
it is still unstained, try a sirius red method (using CI 35780, sirius 
red F3B, NOT sirius red 4B). You might also want to consider thioflavine 
T and the other methods using dyes other than direct cotton dyes.

We do all of our Congo Red staining on Muscle and Nerve frozen sections.  
Sometimes when there is very little amyloid detected using the 
bright-field/polarizing microscope our pathologist will confirm under a 
fluorescent microscope with the Rhodamine Red filter.  This shows the amyloid 
inclusions very well.

This may or may not be of help - Millipore's xAPP (MAB348) works on horses;
HIER using high pH retrieval solution.  I don't have data on reactivity on
pigs yet.

Does anyone out there in Histoland have experience with this antibody? I have 
been asked to look into running IHC on pig brain on the Bond Max platform. I am 
looking into the antibodies from LS Bio and Invitrogen/LifeSciences, and would

Drew,

At one time we were going to bring this stain in-house (Amyloid A) but
finding positive control material was problematic.  One of our
pathologists remembered a case from more than 20 years ago (and had
documented it!) but it was still just a tiny renal biopsy.  We found
that ProPath (214-237-1894, www.propath.com ) does this antibody so we
gave them some of our renal biopsy and since then have sent our cases to
them for staining.

[Histonet] Amyloid A- IHC stain

Hello fellow Histonetters,

I was wondering if anyone is running Biocare's Amyloid A or Amyloid P 
antibodies on Ventana's Benchmark. If so, are you using the concentrated or 
predilute antibody?

Also, are you running these alone or with a Congo Red stain (as in a panel)? 
Any information is greatly appreciated.

Dorothy,

We've done several comparisons like this for amyloid staining. In our
experience, 8um works fine. We did see stronger staining (birefringence) in
some thicker sections with Congo Red but I don't think there would be a
significant improvement in staining (sensitivity?) from 8um to 10um. I
wouldn't cut sections any thinner than 8um but you can still get amyloid
staining at 4um. It would be relatively easy to take your controls and cut
them different thicknesses just to prove the point. Good luck!
