Hi all,

I do have MAC387 and it works great in dog and pig tissue.  The PI is
asking for additional CD markers to macrophages (CD68 and 204).  I need
them to work on dogs and pigs, but of course, there's not many vendors who
list species cross-reactivities on their data sheets beyond the usual human
and mouse.

In my earlier trials years ago with CD68 (clone KP1), I didn't have success
using it on dog tissue.  Am looking for a different clone that will
cross-react with dogs.

Hi,

MAC387 is a great marker but it detects less macrophages than CD68. In 
our experience (sheep and cattle), MAC387 is good for labelling 
macrophages that have recently reach the lesion, while those resident or 
long standing macrophages would be negative to MAC387 (but mostly 
positive to CD68, which is a more general marker for macs). No idea 
about CD204 though.

Well, you're going to have dendritic cells staining with CD68.  Dendritic
cells be really large, full of cytoplasm, and multinucleated.

I guess that brings you back to "hopefully macrophages".  Maybe someone can
recommend a more specific marker or better criteria for what a macrophage
is...

[Histonet] CD68 counts

I am attempting to accurately assess the number of CD 68 positive cells 
(hopefully macrophages) in biopsies,  they have been GMA processed, ABC 
staining, with AEC visualisation;  no problem with a nucleated cell with 
positive cell  membrane, but  they  are  in  the minority, so  the  question 
how  does one judge the cut-off point that makes a positive cell  into a 
negative cell  if you catch my drift??. I am quite experienced at counting 
cells but feel that obtaining reliable, consistent results from this material 
is quite challenging.
   All ideas/thoughts gratefully received
