I am seeing this when I work with mouse tissue. After necropsy I extract the tissue from the mouse, cassette it, put it in fresh 10% NBF and let the tissue fix 24hrs before processing. I also see an edge effect when I fix with 4% PFA. During IHC-P I usually retrieve with a pressure cooker (120C)
