Does any one have data for the time it takes for methacarn fixative (60%
methanol, 30% chloroform, 10% glacial acetic acid) to penetrate and then
fully fix tissue?

Could anyone give me the concentration (? 0.1-0.05%) and the exact
technique/protocol to use. I am staining shoot apical meristems not leaf or
root.

(small 1 by 1mm)-any advice or modern references as to fixation,
permeabilization, best stain (proabably propridium iodide).

which will end up as 4% when diluted with equal amounts of glut.). Please
let me know what you think.

I am using pure formaldehyde with no methanol and unbuffered as a first
fixative. It comes in concentrations from 20% to 4%. From my reading; 1)it
seems the higher strength penetrates faster ?. 2)4% is the minimum strength
to get good penetration ?.
