I guess it also depends on the application he's using to open the file and when it's written by the other (before or after replication).
 
If the file is replicated between the servers before the first user is closing (saving) the file, and the application is able to handle it, it will inform the user that there's a different version of the file on the server and offer him to reload. But apps which are doing this are pretty rare.

If running DFS on R2 the last write wins, but the first write is put into the Conflict and Deleted folder on the server, so that it can be retrieved if necessary, depending on available space, quotas, etc.
