I'm afraid that's not enough details to provide real help...
Did you check the incl/excl list?
Is there a domain-statement to backup a filesystem that is not a
mountpoint (-> use virtualmountpoint)?

Tried to back up 100 GB file on AIX box.
TSM server is Windows, TSM 5.3.2.
No error in TSM activity log, non on AIX box either.
