Please checkout the 
AdGropuBidModifierService<https://developers.google.com/adwords/api/docs/reference/v201402/AdGroupBidModifierService>and
 the 
AdGroupBidModifer<https://developers.google.com/adwords/api/docs/reference/v201402/AdGroupBidModifierService.AdGroupBidModifier>
.

You may be able to add negative criterions on a campaign by using the 
CampaignCriterionService<https://developers.google.com/adwords/api/docs/reference/v201402/CampaignCriterionService>.
 
 That being said, the ability to target is at best the same capability of 
the AdWords UI.
