A restart is necessary, like with any other change to the rules
or other .cf files. These files are only read once by a parent
process during startup, at that time SA initializes.

No other changes with amavisd are necessary, SA will notice
a directory /var/lib/spamassassin/spamassassin/$VERSION by itself.
