Kevin, Once you have received your certificate, you can upload QSOs in two ways. You can enter them manually with the TQSL software (that's what I do) or, if you are using a compatible logging program (if it generates Cabrillo files, it is), you can do it automatically by uploading the Cabrillo file as a .tq8. If the other station uploads a matching QSO, it will produce a QSL in LOTW.
