You've read the paper. So, you'll know that extreme FIRMA scores (i.e. large positive/negative) represent putative differential splicing events. So, in general, you are looking for large (in magnitude) values. If you have replicates, maybe you want to look for significant changes in FIRMA scores between groups. Hope that helps.
