Create a filter, checking if the value is null and the run if criteria for the filter is true, give an errormessage: for example:
 
('Field1' = $NULL$) and ('Field2' = "X" and Assigned = $USER$)
 
This will make Fieds 1 required in this case, else not.

You need to be aware that every time someone modifies that ticket, they will need to have a value in that field.  With workflow, you can have other scenarios…
