Sounds like a permission issue with the user account that the IIS worker process for that site.
Check which application pool that site is using, then have a look at what account its using. Check the account has write permissions on the windows\temp folder and the asp.net temp fodler in the windows\microsot.net\versionNumber\ASP Temp Files directory. Also the accounts must have a few other permissions (NETWORK SERVICE account has these other permissions by default), not sure off the top of my head, google it. Also, restart the IIS server, then hit a asp.net page on the site, then check the event log, if there's anything stopping the worker process from starting it will appear in the event log.
