For getting started, one thing most people are confused about is how the game saves. Your game wont save until you prestige and start over, then some of your bonuses will carry over to the next play through, and you'll be able to get further and further with each successive session. I would plan each session out around when you are going to prestige so you can progress each time. The other thing is make sure you start on easy, then move to medium and hard when you have enough prestige bonuses and heirloom gear. If it gets too hard, go back to the previous difficulty for a while.
