I believe it may be possible to dynamically add configuration for a block but I don't think it is currently possible to add a block to assembly.xml. It is a direction we are planning to go but we aren't quite there yet ;)
In the meantime - you may want to experiment with the Merlin container (excalibur/assembly).
It provides support for dynamic component addition.
