This I have now done. The only problem is that there doesn't seem to be any braille output. Looking in the logs, brltty is syncing with polkit but polkit might be denying me access, and I'm the only user logged in. This is after a reboot. I've uncommented the line that configures brltty to use polkit, is there anything else I need to do?

That should generally work fine. An alternative, if you have brltty 5.4, would be to specify api-parameters auth=polkit in brltty.conf. This will tell brltty to let polkit decide whether the user should have access to the Braille display, rather than using brlapi.key (the default policy gives access to the user logged in locally but not to a user logged in remotely).
