I think there are missing environment variables. Most of us have a script that sets them on Linux, and on Windows, you have to set some environment variables in your profile.
I don't think we open system generator from the start menu, generally. I take it you are trying to use it outside of the casper toolflow? In that case, I think your best bet is the Xilinx web site for info.
