Hello All,
I wonder if anyone has experience with making semet substituted proteins in insect cells. Specifically, we have been making native protein using sf21 cells grown on Sf900 II media from invitrogen.

I see that invitrogen also carries Sf900 II media that lacks methionine and cystine. My experience with other media preparations (I've used Excel 405 before) was that for best results I had to special order it without methionine and also without yeastolate. I have been unable to find out from Invitrogen whether Sf900 II contains yeastolate (ingredients are secret; also, I was told that special ordering would cost 3K in USD for 6L, and I am still coughing and not sure about coughing up 3K). so I wonder if anyone has made semet substituted protein using sf900 II without met and cystine, as offered by Invitrogen--and whether semet substitution was sufficiently high for SAD/MAD?
