Hi everyone,
I'm encountering problems with silverstaining of 15% Tris-Tricine
Gels. I'm using a standard protocol:
1. Fixation 50% MeOH 12% Acetic acid (1 hour - overnight)
2. Washing in 50% EtOH (3x 10 min)
3. 1min 0,2% Sodiumthiosulfate
4. 3x 20min H20
5. Impregnation 2g/L AgNO3 (+150µL Formaldehyde in 200mL) (20 min I
tried up to 1,5 hours)
6. Washing for around 30 min H20
7. Development Sodium carbonate 60g/L; 100µL Formaldehyde (200mL
solution); 4µL Sodium thiosulfate (of 0,2 g/mL solution)
The results are that I don't get any staining at all using a virgin
gel and a more or less destained Coomassie stained gel.
