Dear all, Recently, I have solved a low resolution structure (2.85 Å) by MR; an alpha-beta mix. In this structure, one of the alpha helices (which is missing in the model) has a tube of density where I can only trace the main chain. In COOT, upon Map sharpening adjustments I could see density for few side chains. In this regard, I have some doubts about B-factors and map sharpening. To my knowledge, refinement of atomic B-factors at lower resolution is not informative and sharpening in COOT can be used only to enhance the resolution details. Is there any approach for integrating or regularizing the B-factors map sharpening in refinement for such a low resolution data?
I hope, am not mixing up different things together. Correct me, if am wrong anywhere and direct me to the references on this. Thank you, Raspudin
