Yes, you can. We have!

We have a 404 handler in Apache which hands off to a specific CF script 
(with the referring URL). We also use the same CF script as a missing 
template handler on CFMX.

In answer to Lee's question, yes, it is site wide but I expect you 
could code a smart 404 template that would pick out the user's domain 
and redirect to their handler.
