The extension system is a work in progress.If I am not mistaking, a preliminary support for extensions exists in the Chrome 3 beta release. But more support (but still really not complete) exists in the Dev release ( dev.chromium.org/getting-involved/dev-channel).
You need to edit the Google Chrome shortcut (right click on it and select properties) and append --enable-extensions to the target.
