This isn't a Cisco issue, but the byproduct of a missing "new" entry in the ARP table. You may know the layer 3 address for this new interface and the router may know how to route it (i.e. this subnet exists off of this interface), but it does not yet have the layer 2 address needed for final delivery. So- your first series of Ping packets defaults to inverse ARP, and if properly configured, the new interface is known.
