Look at clamd.log (or whatever you configured as LogFile).
There should be lines like:

Wed Jan 28 14:16:26 2004 -> Reading databases from /var/lib/clamav/
Wed Jan 28 14:16:27 2004 -> Database correctly reloaded (20583 viruses)
