This may be produced by a corrupt desktop file. To rebuild it, boot the machine normally from the internal hard disk and when the happy Mac icon is shown, press Command and Option and keep them pressed until an alert is shown asking for confirmation to rebuild the desktop. Then you can release both keys and click the "Yes" button. Take into account that you may have to press Command+Option for several tens of seconds, or even for more than a minute (depending on the boot tiem of your machine), so be patient. Also take into account that rebuilding the desktop with a 'large' hard disk (like yours) can take several minutes. If all goes well, you'll be able to boot into the Finder.
