I have built a DNA into a cryoEM map and I want to change the directionality of the chain. Is there a simple Coot option that I can use or do I have to build the model again.

Not that I can think of.

Another question is: can Coot add DNA in both directions, 5' and 3 ',

Yes.

if so, how to reverse the directionality while baton building.

Baton building is for proteins. It will place CAs based on the expected density distribution of peptides. I'd be surprised if it can usefully be used for DNA.

If I were building DNA, I'd first try nautilus or cootilus and then edit using residue by residue extension as needed.

Paul.
