What is your DEFAULTDELIVERY setting in your courierd config file? It should be DEFAULTDELIVERY="| /usr/local/bin/maildrop" if maildrop is to be used for local mail delivery. Or, if you like it only for some mailboxes, use .courier file to enable it.
