Dear Rosea, ideally, your package would be built for Debian in a Debian chroot, and for Ubuntu in an Ubuntu chroot, which would contain sources.list files restricted to the strict minimum. This way, not only it makes sure that the package does not need accidentally some software that is not packaged, but it also ensures that the dependancies are set correctly in the binary packages for the very distribution that you are targetting. While you can do this on chroots that you manage by hand, there are two popular tools for this purpose: sbuild and (p|cow)builder. There is plenty of doc about them on the interweb.
