Could you please run Wireshark on the server listening on the outgoing
route's interface, then restart apt-cacher-ng and retry that failing
download attempt? And then save the pcap log and send it to me.

That's another bug happening when releasing previous file item. Thanks
for discovering it. It is not related to your original issue but it
prevents the original file descriptor from being destroyed, and
therefore when the error above happened once then the proxy will
continue reporting the same error message instead of trying another
download.
