AFAIK, dpkg-dev is only invoked at source-unpack and package-build time, i.e. you need a full DEBIAN/ directory for dpkg-dev to act on. At that time, either you have thrown away the debugging information already (via dh_strip, e.g.), or not. This is all AIUI.
