If you mean /etc/fonts/fonts.conf -- it says:

DO NOT EDIT THIS FILE.
IT WILL BE REPLACED WHEN FONTCONFIG IS UPDATED.
LOCAL CHANGES BELONG IN 'local.conf'


Whether that means:
/etc/fonts/conf.avail/51-local.conf
or
/etc/fonts/conf.d/51-local.conf

/etc/fonts/conf.d/51-local.conf alludes to a local.conf file:
        <include ignore_missing="yes">local.conf</include>

So you make changes in /etc/fonts/local.conf or
/etc/fonts/conf.d/local.conf ?

It all seems rather convoluted. :-(

No doubt, spending hours searching for the appropriate documentation
might help.
