I created a newuser as requested. Trying to play the movie produced the
same results, it did not play and no message as to why.

I also tried running totem from the command line, no additional messages seen.
I also tried running totem in dedug mode, no messages that appeared seemed to 
relate to the issue:

$ totem --debug

After your comment that "that's more like a codecs issue and not a totem
bug" I remembered that I had previously installed the package (via
synaptic): ubuntu-restricted-extras

I removed this package (via synaptic).
I still got the same response when trying to load the mp4 file, it did not play 
and no response at all as to why. Even after a reboot.

The file I am trying to play does appear in the "Supported files" list
when I was selecting it from within totem, both with and without the
"ubuntu-restricted-extras" packages installed.
