Using DirectFB as a replacement for X is currently not possible for
several reasons.

The multi-application core that allows to run mutiple DirectFB
applications simultaniously is not stable yet and there are not enough
applications availible to make up a reasonable desktop (i.e no gnome :-)
). since gtk-2.0 applications run in DirectFB natively this situation
might change in the future..

Another way is to use the XDirectFB server. This doesnt need the multi
application core and can be uses as a replacement for your regular X
server. You will get some extra features such as transparent X windows (
http://www.directfb.org/screenshots/index.xml ). Some people I know
already use this.

The ultimate goal is to use the multi application core with as many
native DirectFB applications as possible and start XDirectFB as one
DirectFB process, which will allow you to start "legacy" X applications
nicely integrated in your DirectFB desktop. This is possible today, but
not stable yet.
