I'm having this same problem since going to 6.5.x. (Currently running 6.5.1 nightly from last week, WindowsXP, SB3).
Never ever happened before. Symptoms are as described, cannot wake slimserver by just turning on SB3, must repeat several times. Other symptom is that from PC/server, when I 'start slimserver'...my player does not show up, so I cannot just start playing music from my PC/server (whereas before I could). I have to go over and turn the SB3 on (and off and on...) again.
