I just use Cool Edit pro.
if I have the film on DVD I can set my sound card to record CD audio and
just set Cool Edit to record while I start the film off at the right point.
once the sample has finished, stop recording and trim the wave down so that
you end up with just waht you want.

There are several web sites out there that have samples from films, so if
you nkow what you are lokoign for you could download it.
