I am planning to test the germination capacity of seedbanks from Low Arctic tundra and am looking for the best methods for broad-spectrum germination. The literature reports mixed results with samples stratified at 0-4C for a number of weeks vs samples incubated directly from thawed frozen soils. Almost all methods resulted in more graminoid germination than dicots which might be due to more graminoid seeds present in the soils, but which in part might also be due to dicots being more difficult to germinate. I would be interested to hear what others have tried?
Thanks very much! ~Diane Huebner
