One difference regardless of what transaction you are using is in the Interchange ISA segment element-11. This element represented the Interchange Control standard ID which was always a 'U'. After version 00410, the element is used to represent a repetition separator.

Your best bet would be to do a websearch on the differences or go to the "source" - the group that created and manages the specifications. Chances are that there may be no changes between the 4010 and 5010 versions - depending on the documents and segments you're going to be using. But then again, it could be that there are MANY differences between the versions - again, based upon the documents and segments you will be using.
