The SMTP port has nothing to do with the error you're getting if you're using the Outlook client in Workgroup/Corporate mode. Since you've got an Exchange Server, I'll assume that this is the case.
When you say you can ping it, you're sure that you can ping it by name, right? Are you logged on to the computer with those credentials? Can you send mails to that mailbox? What happens when you logon as another account and try to open the Inbox for this mailbox using Open Other User's folder?
