Are you using Mailbox Manager (comes with SP3 / SP4)?
Useful for cleaning out deleted and sent items.

One thing we found here which was chewing up private IS space
was journals.  If your users are using them, do they need to?
If they do, how long do they need to retain them?  I've
configured Mailbox Manager here to delete old journals
automatically.  When I did that, we freed up 3GB of IS space.
