Is there an ELB (load balencer) or VPC involved ? If so they have their own set of ACL's Its also conceivable the EC2 instance got updated and iptables changed (or your desktop). Suggest trying to connect form a different client host to eliminate possible local firewalls as a problem. Next I would ssh to the instance and use telnet (or curl or wget) to localhost:8001, and check iptables to see if the server itself is blocking the port or if its from 'outside'
