Hi Deqiang, I cannot access your example files (I assume you are using these on a local mirror), so I tried to reproduce this myself.
I just confirmed that gene models (bed 12) work correctly for bigBeds. Please change: type bigBed to type bigBed 12 in your hub trackDb file. Colors do display correctly in bigBeds. If you are using colors, you have to include: itemRgb on in your trackDb for the track. See details here (in example three): http://genome.ucsc.edu/goldenPath/help/bigBed.html and here: http://genome.ucsc.edu/FAQ/FAQformat.html I think you will have to got to the track hub page and click on "Load Selected Hubs" to reload the hub configuration files after you have modified them. Also make sure your track is not showing in "dense" mode. To check this, right-click on the track and change to "pack" or "full" to see track details (including gene structure).
