In my limited experience, even with a not-so-exciting storage solution, file I/O has not been a limiting factor in rendering raster in GeoServer. So we store everything-- overviews too-- uncompressed. I know others apply jpg compression to their overviews, but leave the raw stuff uncompressed. Now PostGIS I/O (for multi-gigabyte geometries) , well that's a different story. :)
